Engineering of a Peptide ??N?Methyltransferase to Methylate Non?Proteinogenic Amino Acids

چکیده

Introduction of ?-N-methylated non-proteinogenic amino acids into peptides can improve their biological activities, membrane permeability and proteolytic stability. This is commonly achieved, in nature the lab, by assembling pre-methylated acids. The more appealing route methylating amide bonds challenging. Biology has evolved an ?-N-automethylating enzyme, OphMA, which acts on fused to its C-terminus. Due ribosomal biosynthesis substrate, activity this enzyme towards with not been addressed. An engineered intein-mediated protein ligation solid-phase peptide synthesis have allowed us demonstrate methylation context non-natural amides. approach may application biotechnological production therapeutic peptides.

In Vitro Mutasynthesis of Lantibiotic Analogues Containing Nonproteinogenic Amino Acids

Lantibiotics are ribosomally synthesized and post-translationally modified peptide antibiotics containing the characteristic thioether cross-links lanthionine and methyllanthionine. To date, no analogues of lantibiotics that contain nonproteinogenic amino acids have been reported. In this study, in vitro-reconstituted lacticin 481 synthetase was used in conjunction with synthetic peptide substr...

Non-standard amino acids in peptide design and protein engineering

The introduction of non-coded amino acids with well defined stereochemical and functional properties will greatly enhance the scope of protein design and engineering. The present state of methodologies for incorporation of non-coded residues into proteins is examined. The prospects for conformationally constrained amino acid residues are evaluated in the light of peptide structural studies. Tem...

Energetics of side-chain snorkeling in transmembrane helices probed by nonproteinogenic amino acids.

Cotranslational translocon-mediated insertion of membrane proteins into the endoplasmic reticulum is a key process in membrane protein biogenesis. Although the mechanism is understood in outline, quantitative data on the energetics of the process is scarce. Here, we have measured the effect on membrane integration efficiency of nonproteinogenic analogs of the positively charged amino acids argi...

Design of supramolecular amino acids to template peptide folding.

We describe in this manuscript a new design of supramolecular amino acids (SAAs) for directing peptide folding in organic environments. The incorporated supramolecular motif has a strong driving force to dimerize in a sequence- and orientation-specific manner. By introducing such SAAs into the primary sequence of peptides, the specific and directional dimerization of the supramolecular units sh...
